It doesn't really matter. I would get in a routine and do it in the morning.

I like to poke first thing in the morning.I have tried poking in the evening,it seems to make it harder to sleep that way.

As soon as the syringe is full ;)
I like popping at night, so it has some time to absorb before the next day's lifting. (Short esters)
But as said before, be regular about it, and do what is most convenient for your schedule.

I like to "poke" in the morning before I eat and after I take a shower! SO my muscles are somewhat relaxed!
