Excitement over weight loss drug
Could a human version be produced?
Monkeys given a new hormone-like compound lost a massive seven per cent of their body weight - in just one week. Researchers are now hopeful that a drug can eventually be developed to tackle the "obesity epidemic" threatening the West.
The compound, called KB-141, works on the same principle as the hormone produced by the thyroid gland in the neck.
It is well known that giving this to patients can produce weight loss by boosting the metabolic rate - so that the body burns more calories.
However, this comes at a price, as the hormone also causes an potentially-dangerous increase in the heart rate.
Scientists hoped that if they could somehow separate the two effects, then perhaps a drug could be made which would produce the weight loss without the heart problems.
A team of researchers from the Karolinska Institute in Sweden and the University of California, in San Francisco, believe they have found a way to do this.
Selective effect
Thyroid hormone works by stimulating two main receptors on the body's cells.
One of these, called TR-alpha, appears to control heart rate, while the other, TR-beta, appears to regulate the metabolism.
They have developed an experimental chemical called KB-141 which they believe stimulates TR-beta, but not TR-alpha.
Animals given the drug showed decreased cholesterol levels and reduced body weight, but crucially, without a faster heartbeat.
When the compound was given to monkeys, there was a seven per cent drop in body weight over just seven days - with no effect on heart rate.
"Our results in three species show that a separation between potential beneficial versus deleterious effects is possible," say the researchers, writing in the Proceedings of the National Academy of Sciences.
Obese rise
A drug which could control obesity is a "Holy Grail" for researchers, as record levels of the disease are recorded in many western countries.
The research needs to proceed with caution - with longer term studies, probably in monkeys

Drug companies are racing to find ways to stimulate the body to shed weight, with the promise of vast profits for a successful compound.
Other areas of research include "gene therapies" designed to tackle genetic mutations that predispose to obesity, and drugs designed to reduce hunger.
Professor John Lazarus, the president of the British Thyroid Association, said that the finding was "very exciting".
He said that the effects on the heart meant that it was dangerous to give thyroid hormone supplementation to patients unless it was clear their own gland was not making enough.
"This certainly shows promise - the amount of weight loss is impressive, although there are question marks over whether this loss could be sustained in a human patient.
"The research needs to proceed with caution - with longer term studies, probably in monkeys."
He said that it had to be proven that the new compound avoided another damaging side-effect of getting too much thyroid hormone - bone loss.

hmmm, While I am inclined to jump on the bandwagon and praise this new drug I must admit I have certain reservations when it comes to the "magic pill" as mbaraso so delicately put it.

Errr... maximum of 5 files per post so I split them up.

Few treatments for obesity exist and, whereas efficacious therapeutics for hyperlipidemia are available, further improvements are desirable. Thyroid hormone receptors (TRs) regulate both body weight and cholesterol levels. However, thyroid hormones also have deleterious effects, particularly on the heart. The TR beta subtype is involved in cholesterol lowering and possibly elevating metabolic rate, whereas TR alpha appears to be more important for control of heart rate (HR). In the current studies, we examined the effect of TR beta activation on metabolic rate and HR with either TR alpha 1-/- mice or the selective TR beta agonist KB-141 in mice, rats, and monkeys. 3,5,3'-triiodi-l-thyronine (T3) had a greater effect on increasing HR in WT than in TR alpha-/- mice (ED15 values of 34 and 469 nmol/kg/day, respectively). T3 increased metabolic rate [whole body oxygen consumption (MVO2)] in both WT and TR alpha-/- mice, but the effect in the TR alpha 1-/- mice at the highest dose was half that of the WT mice. Thus, stimulation of MVO2 is likely due to both TR alpha and -beta. T3 had equivalent potency for cholesterol reduction in WT and TR alpha-/- mice. KB-141 increased MVO2 with selectivities of 16.5- and 11.2-fold vs. HR in WT and TR alpha 1-/- mice, respectively. KB-141 also increased MVO2 with a 10-fold selectivity and lowered cholesterol with a 27-fold selectivity vs. HR in rats. In primates, KB-141 caused significant cholesterol, lipoprotein (a), and body-weight reduction (up to 7% after 1 wk) with no effect on HR. TR beta-selective agonists may constitute a previously uncharacterized class of drugs to treat obesity, hypercholesterolemia, and elevated lipoprotein (a).
Now the above study kb-141 vs T3 for heart rate, fat loss and cholesterol lowering.
KB-141 basicly caused mad fat loss WITHOUT the nasty T3 sides increased heart rate etc, this is because KB-141 binds to the receptor to promote fat burning but not the one to control heart rate! It's like taking all of the positives of T3 and none of the negatives, cool stuff.

did some googling for KB-141...looks like theres no human version out as of yet...if this comes out it'll be a major breakthrough like the viagra pills were a couple of years ago....
