I know this will vary between individuals, but if one had a choice of running 400mg or 500mg of test prop each week, would you notice that much of a difference as far as gains go?

all depends on training and diet, by theroticaly if you had two identical twins who ate and trainend and did everything exatly the same the one on 500 should see more gains, i mean it is 500mg which is a fair bit its like 25% more then 400mg. Would you see much diffenence i dont know but its 100mg so why not.
