Wear cycling shorts

She is 300lbs and about 5feet tall. They don't make shorts that big I don't think lol.

2 suggestions :
1- gel seat cover - easily avail and cheap
2- If that wont cut it -depending on type of spin bike/how seat is mounted- it is likely that an exercise bike seat can be mounted in place of the regular saddle on the spin bike.
