Blood Test Timing for E2

Any advice on the best timing for a blood test for E2 in relation to AI intake?

I want to determine what the ideal AI dosage should be going forward but I have already started taking an AI today. Today I took my first dose of a little over 0.25mg anastrozole. Should I get my E2 tested today, tomorrow, or the day after tomorrow before my next dose? Should I wait altogether for a week or so on 0.25mg EOD?

I am on 300mg test prop per week + 500iu HCG
(and also 60mg oxandrolone ED but oxan doesn't aromatize)
Pre-cycle my E2 was at 26 pg/mL

I was also hoping to keep an eye on my hematocrit. You really think waiting 6 weeks before a blood test is okay? My hematorcit was at 43,3% pre cycle.

I don't know how long I can cycle for, I originally planned 6 weeks with oxan, now I want to extend and was considering 8 weeks. I've heard also that with testosterone it should be a 12 week cycle. If I do only 6 weeks, waiting until the end of the cycle seems pointless, if I extend to 12 weeks it would be the half-way point so it makes sense in a way.
